Research Interest:

My research interest is Epigenetic Regulation in Cancer Biology, with the focuses on functional studies of novel oncogenes and tumor suppressors. Our work suggested an oncogenic role of Yin Yang 1 (YY1) through negatively regulating p53 and p27 tumor suppressors and the regulation is independent of YY1’s transcriptional activity. I invented the DNA vector-based RNAi vector (shRNA), the first generation of pSilencer (a product of Ambion, Inc.). The PNAS paper for this approach has been cited over 1,000 times and this technology has become a common technique in most research labs. We are also investigating functional roles of other tumor suppressive proteins and oncogenes, including noncoding RNAs, in cancers.

Our current research focuses include (1) Molecular mechanism for the anticancer activities of plant compounds and their combinatorial use with chemotherapeutics in cancer treatment. (2) G-quadruplex: formation, function and regulatory mechanism. (3) Molecular mechanism of YY1-mediated enhancer activity in regulating gene expression. (4) Tumor suppressors and oncogenes: molecular mechanisms and potential targeting sites for cancer therapies.
